Serum Biomarker Discovery: Discovery Means Finding Things You Didn’t Expect

Resumen

We have developed a robust and reproducible way to identify biomarkers for human diseases within the blood serum, using discovery-based mass spectrometry of large-protein-depleted specimens.  We have applied the method to complications of pregnancy—including preterm birth and preeclampsia—and to Alzheimer’s disease, finding several markers for each of these conditions.  Though the method was developed to identify biomarker candidates among the low-molecular-weight serum proteome, lipids were not excluded during the preparation and many lipidomic biomarkers were subsequently observed.  Our method shows tremendous promise for identification of disease risk before the onset of clinical symptoms.  Additionally, the importance of lipidomics in preclinical characterization is underscored.
